Fourier flim multiplexing (FmFLIM) allows multiplexing tomography 3D imaging lifetime of the entire embryo. In FmFLIM our previous system, the spatial resolution is limited to 25 pM as a trade-off between spatial resolution and imaging depth. In order to achieve cellular resolution imaging of thick specimens, we build tomography system with dual-color Bessel beams.
In combination with FmFLIM, Bessel FmFLIM tomography system can perform parallel lifetime imaging 3D on channel excitation-emission couple on cellular resolution of 2.8 um. Ability Bessel tomographic image of the system is shown by 3D imaging FmFLIM lifetime of dual-labeled transgenic zebrafish embryos.
